This standard specifies the liquid chromatography-tandem mass spectrometry method for the determination of papaverine, morphine, nicotine, codeine, and tibaine in hot pot food. This standard is applicable to the determination of papaverine, morphine, nicotine, codeine, and tibaine in hot pot foods such as sauces, soups, seasoning oils, and solid seasoning powders. 2. The sample is evenly dispersed in water or hydrochloric acid solution, extracted with acetonitrile, and then salted out and layered. The acetonitrile extract is purified with a bonded silicon solid-phase extraction adsorbent, centrifuged, and detected by liquid chromatography-tandem mass spectrometry. Papaverine, narcodine, and tibaine are quantified using external standard method, while morphine and codeine are quantified using internal standard method. Unless otherwise specified, all reagents and materials used in this method are analytical pure or above, and the water is Grade 1 water as specified in GB/T 6682. 3.1 Methanol (CH, OH: chromatographically pure). 3.2 Acetonitrile (CH: CN): chromatographically pure. 3.3 Formic acid (HCOOH): Chromatically pure. 3.4 Hydrochloric acid (HCI) and 3.5 ammonium formate (HCOONH,). 3.6 Sodium hydroxide (NaOH). 3.7 Anhydrous magnesium sulfate (MgSO) is ground and dried in a muffle furnace at 500 ℃ for 5 hours. At 200 ℃, it is taken out and bottled, stored in a dryer, cooled for later use. 3.8 Anhydrous sodium acetate (CHCOONa) -3.9 ethylenediamine-N-propylsilane (PSA) packing: particle size 40-70 um.3.10 C18 packing: particle size 40-50 um3.11 Hydrochloric acid solution (0.1 mol/L): Measure 9 mL of hydrochloric acid (3.4), add water to 1L, shake well and set aside. 3.12 Ammonium formate solution (10 mmolL): Accurately weigh 1.26 g of ammonium formate (3.5) and dissolve it in an appropriate amount of water, bring to a constant volume of 2 L, mix well, and set aside. 3.13 Formic acid methanol solution (0.5%): Take 1 mL of formic acid (3.3) and place it in a 200 mL volumetric flask. Dilute with methanol and bring to volume. Shake well for later use. 3.14 Sodium hydroxide solution (1 mol/L): Accurately weigh 40 g of sodium hydroxide (3.6) and dissolve it in an appropriate amount of water, bring to a constant volume of 1 L, mix well, and set aside. 3.15 Papaverine hydrochloride, morphine, narcodine, codeine phosphate, and tibaine standards: purity not less than 98%. 3.16 Internal standard substances: morphine D3, codeine D3
