Food additive - Acetylated starch phosphate ester detection

This standard specifies the spectrophotometric method and inductively coupled plasma emission spectroscopy method for the determination of phosphorus content in food. The first and third methods of this standard are applicable to the determination of phosphorus in various types of food, while the second method is applicable to the determination of phosphorus in infant food and dairy products. After digestion of the 2-principle sample, phosphorus combines with ammonium molybdate under acidic conditions to form ammonium phosphomolybdate. This compound is reduced to a blue compound molybdenum blue by hydroquinone, sodium sulfite, or stannous chloride, or peroneal sulfate. The absorbance value of Key Blue at 660 nm is directly proportional to the concentration of phosphorus. Measure the absorbance of the sample solution using a spectrophotometer and compare it quantitatively with the standard series. Unless otherwise specified, all reagents and materials used in this method are analytical pure, and the water is Grade III water according to GB/T 6682. 3.1 Reagent 3.1.1 Sulfuric acid (H, SO,): premium grade pure. 3.1.2 Perchlorate acid (HCIO,): premium grade pure. 3.1.3 Nitric acid (HNO.): premium grade pure. 3.1.4 Hydrochloric acid (HC: superior purity). 3.1.5 Hydroquinone (C, H. O:). 3.1.6 Anhydrous sodium sulfite (NagSO,). 3.1.7 Ammonium molybdate [(NH,), Mo, O, · 4HgO]. 3.1.8 Stannous chloride (SnClg · 2HO). 3.1.9 Sulfuric acid volume (NHNH. · H SO,). 3.2 Preparation of Reagents 3.2.1- Sulfuric Acid Solution (15%): Measure 15 mL of sulfuric acid and slowly add it to 80 mL of water. After cooling, dilute with water to 100 mL and mix well. 3.2.2 Sulfuric Acid Solution (5%): Measure 5 mL of sulfuric acid and slowly add it to 90 mL of water. After cooling, dilute with water to 100 mL and mix well. 3.2.3 Sulfuric acid solution (3%): Take 3 mL of sulfuric acid and slowly add it to 90 mL of water. After cooling, dilute with water to 100 mL and mix well. 3.2.4 Hydrochloric acid solution (1+1): Measure 500 mL of hydrochloric acid, add 500 mL of water, and mix well. 3.2.5: Ammonium molybdate solution (50 g/1.): Weigh 5 g of ammonium molybdate, dissolve in sulfuric acid solution (15%), dilute to 100 ml., and mix well. 3.2.6 Hydroquinone solution (5 g/L): Weigh 0.5 g of hydroquinone in 100 mL of water, dissolve it, add one drop of sulfuric acid, and mix well. 3.2.7 Sodium sulfite solution (200 g/L): Weigh 20 g of anhydrous sodium sulfite and dissolve it in 100 mL of water, and mix well.
