This standard specifies a high performance liquid chromatography/tandem mass spectrometry method for the determination of residues of nitrofuran drug metabolites 3-amino-2-oxalidinone (AOZ), 5-methyl-3-amino-2-oxalidinone (AMOZ), 1-aminohydantoin (AHD), and semicarbazide (SEM) in animal derived foods. This standard is applicable to the qualitative confirmation and determination of residual levels of nitrofuran metabolites such as 3-amino-2-oxazolone, 5-morpholinemethyl-3-nitro-2-oxazolone, 1-aminohydantoin, and aminourea in muscles, viscera, fish, shrimp, eggs, milk, honey, and casings. 2 Normative References: The clauses in the following documents become clauses of this standard through reference in this standard. The principle sample is hydrolyzed with hydrochloric acid, derived from o-nitrobenzaldehyde overnight, adjusted to pH 7.4, extracted with ethyl acetate, and purified with n-hexane. The analytes were qualitatively detected using high-performance liquid chromatography/tandem mass spectrometry, and quantitatively determined using stable isotope internal standard method. Unless otherwise specified, all reagents and materials are analytical pure, and the water is Grade 1 water according to GB/T 6682. 4.1 Methanol: High performance liquid chromatography grade. 4.2 Acetonitrile; High performance liquid chromatography grade. 4.3 Ethyl acetate; High performance liquid chromatography grade. 4.4 n-hexane: High performance liquid chromatography grade. 4.5 Concentrated hydrochloric acid. 4.6 Sodium hydroxide. 4.7 Formic acid; High efficiency liquid phase color harmonic level. 4.8 o-Nitrobenzaldehyde. 4.9 Potassium phosphate trihydrate. 4.10 Ammonium Acetate. 4.11 0.2 mol/L hydrochloric acid solution; Accurately measure 17 mL of concentrated hydrochloric acid (4.5) and bring to volume with water to 1 L. 4.12 2.0 mol/L sodium hydroxide solution: Accurately weigh 80 g of sodium hydroxide (4.6), dissolve in water, and bring to volume to] L. 4.13 0.1 mol/L ortho nitrobenzaldehyde solution: Accurately weigh 1.5 g of ortho nitrobenzaldehyde (4.8). Dissolve in methamphetamine and bring to a constant volume of 100 mL. 4.14 0.3 mol/L potassium phosphate solution: Accurately weigh 79.893 g of potassium phosphate trihydrate (4.9), dissolve in water, and bring to volume to 1 L
