Testing of public places and their supplies

This part of GB/T 18204 specifies the sampling and determination methods for the total number of bacteria, total number of fungi, coliform bacteria, Staphylococcus aureus, and Streptococcus hemolyticus in public places. This section is applicable to the determination of total bacterial count, total fungal count, coliform group, Staphylococcus aureus, and hemolytic streptococcus in public places. Other places can refer to it for implementation. The following terms and definitions apply to this document

2.1 Total bacterial count Public goods and tools have been sampled and processed, On nutrient agar medium at 35 ℃~37 ℃ The total number of mesophilic aerobic and facultative anaerobic colonies grown and developed at 37 ℃ and 48 h. 2.2 Coliforms can ferment lactose, produce acid, gas, aerobic, and facultative anaerobic Gram negative non spore forming bacteria at 37 ℃ and 24 h. 2.3 Staphylococcus aureus grows well on Baird Parker medium or blood plate medium, decomposes mannitol to produce acid, and plasma coagulase positive Gram positive grape like bacteria Cocci. 2.4 Total Fungi Count The total number of colonies formed by culturing at 25 ℃ -28 ℃ for 3 to 7 days on Bengal Red or Sabouraud agar medium. Streptococcus hemolyticus belongs to the genus Streptococcus and is a Gram positive bacterium. It decomposes glucose, produces acid but not gas, and produces hemolytic circles on the blood plate. 3. Plate counting method for total bacterial count 3.1. Culture medium and reagents 3.1.1. Composition of physiological saline: 8.5 g of sodium chloride in distilled water l 000 mL Preparation method: Weigh 8.5 g of sodium chloride and dissolve it in 1000 mL of distilled water. Divide into test tubes, 10 mL per tube, and sterilize under high pressure at 121 ℃ for 15 minutes
